Bremelanotide is a synthetic cyclic heptapeptide of the melanocortin chemical class. The ring is closed by a lactam bridge between the side chains of an aspartic acid and a lysine residue, with an N-terminal acetylated norleucine outside the ring and a free C-terminal acid. It is the des-amide analogue of melanotan II, differing from that molecule at the C-terminus, and both derive structurally from the His-Phe-Arg-Trp core motif of alpha-melanocyte-stimulating hormone.
The molecular targets are the melanocortin receptors, a family of five class A G-protein-coupled receptor subtypes designated MC1R through MC5R. Subtype affinity is determined in recombinant cell membranes individually expressing each subtype, by competition displacement of a labelled reference ligand, producing a per-subtype Ki profile. Gs coupling is quantified in the matched transfected cell lines as cAMP accumulation. All reported values are properties of the specific recombinant construct, host cell line and buffer system used.

Primary literature on cyclic lactam melanocortin peptide chemistry and on MC1R-MC5R receptor binding and cAMP assay methodology is indexed in PubMed, with structure and identity records catalogued in PubChem and ChEMBL. Search results and cited literature do not establish the performance of a commercial lot or a use recommendation.
It is supplied for laboratory research and analytical work only and is not intended for human or veterinary use.
Each vial is traceable to a per-lot certificate of analysis recording the HPLC purity result, measured content, mass confirmation, testing laboratory and test date for that specific lot.
LC-MS mass confirmation distinguishes the intact lactam-bridged cyclic peptide from the linear ring-opened species, which differ by 18 Da, and the HPLC trace resolves them as separate peaks.
