Retatrutide is a synthetic 39-amino-acid peptide built on a glucose-dependent insulinotropic polypeptide (GIP) backbone, carrying α-aminoisobutyric acid (Aib) substitutions at positions selected for conformational constraint and protease resistance. A C20 diacid lipid chain is attached at a lysine side chain through a γ-glutamate and OEG spacer arrangement. The molecule is a single-chain, C-terminally amidated peptide with one defined acylation site.
Three class B1 G-protein-coupled receptors are the molecular targets characterised in vitro: the GIP receptor (GIPR), the glucagon-like peptide-1 receptor (GLP-1R) and the glucagon receptor (GCGR). Binding affinity at each is determined by competitive displacement against membranes from cells recombinantly expressing a single human receptor. Functional occupancy is quantified separately at each receptor as cAMP accumulation with EC50 values calculated per receptor, and as beta-arrestin recruitment by enzyme-fragment complementation. Comparative potency across the three receptors is expressed only as a ratio of these in-vitro readings.

Primary literature on multi-receptor peptide design, Aib-substituted backbone chemistry and class B1 receptor pharmacology is indexed in PubMed and in peer-reviewed medicinal-chemistry journals, with compound records in PubChem and ChEMBL. Search results and cited literature do not establish the performance of a commercial lot or a use recommendation.
It is supplied for laboratory research and analytical work only and is not intended for human or veterinary use.
Unopened vial: store sealed, dry, and protected from light at controlled room temperature or refrigerated. After opening or reconstitution: keep refrigerated at 2–8 °C.
The per-lot certificate of analysis reports HPLC purity with the chromatogram, mass confirmation against the calculated molecular weight, appearance, and the lot identifier printed on the vial label for traceability.
