Semax is a synthetic heptapeptide consisting of the ACTH(4-7) fragment Met-Glu-His-Phe extended at the C-terminus by the tripeptide Pro-Gly-Pro. The Pro-Gly-Pro extension is a synthetic addition not present in the parent corticotropin sequence. The peptide is unmodified at the N-terminus, contains a single methionine thioether and one histidine imidazole, and is supplied as a linear, non-cyclised chain.

As an ACTH fragment analogue, Semax is examined against the melanocortin receptor family. Binding at MC1R, MC3R, MC4R and MC5R is quantified by radioligand displacement in membrane preparations from transfected cell lines, with affinity reported as Ki or IC50. In cultured cell systems, BDNF and TrkB transcript and protein levels are quantified by qPCR and immunoblot as endpoint analytical readouts. Measurement terminates at receptor-occupancy constants and transcript- or protein-level signal.
