Selank is a synthetic heptapeptide built from the endogenous tetrapeptide tuftsin (Thr-Lys-Pro-Arg) extended at the C-terminus by the tripeptide Pro-Gly-Pro. It carries three proline residues, a free N-terminal threonine and basic lysine and arginine side chains, giving a strongly cationic peptide at neutral pH. The chain is linear and non-cyclised, with no cysteine and no disulfide bond.

Selank is examined against the molecular targets of the tuftsin scaffold, including the neuropilin-1 binding site and GABA-A receptor membrane preparations. Displacement is quantified by radioligand binding in isolated membrane fractions, with affinity expressed as Ki. Enkephalin-degrading enzyme activity is measured in cell-free hydrolysis assays with the peptide present as the added test article and substrate turnover followed by HPLC or fluorimetry. Readouts terminate at binding constants and enzyme rate values.
