Sermorelin is the synthetic 1-29 amino acid N-terminal fragment of human growth hormone-releasing hormone, prepared as the C-terminal amide. The sequence is unmodified relative to the native 44- residue parent peptide across those first 29 residues and retains the methionine at position 27, which accounts for the single sulfur atom in the molecular formula. It is the parent structure from which several substituted GRF(1-29) analogues are derived.

The molecular target is the growth hormone-releasing hormone receptor (GHRHR), a class B G-protein- coupled receptor. Affinity is measured in recombinant GHRHR-expressing membranes by saturation and competition binding against a labelled reference ligand. Gs coupling is quantified in transfected cell lines by cAMP accumulation, typically read by HTRF or luminescent reporter formats.
